Agilent Expression Profiling Service
Introduction
The Agilent microarray platform is among the world´s leading DNA chip technologies. Based on 60mer oligonucleotides and a very flexible ink jet printing device Agilent expression arrays offer many outstanding features:
- Very high dynamic range (>4 logs)
- Very high accuracy, as measured by comparison with TaqMan RT-PCR (r>0,9)
- High reproducibility (CV of signal values <10%)
- Cost-effective multiplex formats (e.g. 4plex, 8plex)
- Low amount of starting material needed (200 ng of total RNA)
Applications
- Comparison of normal vs. diseased tissue e.g. normal vs. tumour, non-infected vs. infected, wild type vs. knock out
- Time courses e.g. developmental stages, environmental changes, clinical treatments
- Test of influence of chemical compounds, e.g. drugs
Service range
- Full Service - customer sends total RNA (1 µg) or mRNA (100 ng):
- Quality control of total RNA
- cDNA synthesis
- cRNA synthesis and labelling
- cRNA purification and quality control
- Fragmentation
- Hybridization, washing, and scanning of appropriate Agilent microarray
- Data extraction with Agilent´s feature extraction software
- Core Service: customer sends non-fragmented, Cy3/Cy5-labelled cRNA (3 µg)
- Quality control of labelled cRNA. Further steps same as Full Service option (5.-7.).
- Additional Services (extra costs apply):
- Array Design with Agilent´s eArray Tool
- Extended Data Analysis with Agilent´s GeneSpring software (determination of differentially regulated genes, hierarchical clustering, principal component analysis, etc.)
Quality control
- Quality and quantity of each RNA sample sent to ATLAS Biolabs will be determined by analysis on an Agilent 2100 Bioanalyzer and by photometric analysis.
- Quantity and Cy3/Cy5-CTP incorporation rate of each cRNA sample synthesized at ATLAS Biolabs will be determined by photometric analysis
- All quality assurance procedures are electronically documented and available for our customers on demand.
Data delivery
- 2–4 weeks after receipt of samples (for >40 samples, please inquire)
- CD-ROM/DVD sent to customer including:
- Original data
- Results file in XLS or TXT format
